pEZSeq Cloning Kits contain: Vector Premix, CloneSmart® DNA Ligase, Positive Control Insert DNA, Sequencing Primers, and a complete protocol. Kits with cells include E. cloni® ELITE 10G or 10GF' (>2 × 1010 cfu/µg) or 10G SUPREME (>4 × 1010 cfu/µg) Electrocompetent Cells; or Chemically Competent cells: 10G (>1 × 109 cfu/µg) or 10GF' (>5 × 108 cfu/µg), packaged in two transformations per tube (DUOs), Recovery Medium, and Control pUC DNA. Kits without cells can be used with any E. coli chemically competent or electrocompetent cells.
Description
Clone quickly and facilitate downstream applications
Lucigen’s unique cloning kits with pre-processed vectors and competent cells eliminate hours of tedious preparation and enable highly efficient cloning. Ligation can be completed in as little as 30 minutes with no time-consuming clean-up needed. Only a brief heat denaturation is necessary before transformation.
The pEZSeq™ vector (Figure 1) is optimized for high efficiency blunt-end cloning and sequencing in a blue/white screening vector. It is the only commercial preparation of a blue/white screening vector that gives 99% recombinant colonies; common pUC based vectors often produce as many as 30-60% non-recombinant blue colonies. The low background of blue colonies with the pEZSeq kit virtually eliminates robotic picking errors and greatly simplifies manual clone selection. The vector has minimal sequence between the cloning site and the primer binding sites, resulting in up to 5-10% more insert sequence. pEZSeq is small (2,056 bp) and contains little non-essential sequence, so unwanted transposition events are minimized. This small vector design facilitates cloning larger inserts and simplifies subsequent manipulations. For example, large inserts can be rapidly sequenced with transposon insertional sequencing.
