Custom Library Screening

Lucigen provides a library screening service to assist clients with identifying BAC and/or fosmid clones from custom BAC/fosmid library constructed at Lucigen or other library supplied by the client. Library screening is performed via hybridization using synthetic oligonucleotide probes (one or more overgo probes) to custom-prepared nylon blots. Each blot is a 22 x 22cm nylon filter with 18,432 BAC/fosmid clones spotted in duplicate. The overgo probe is designed and generated from 24-mers containing an 8-base pair overlap. The probe is made radioactive in a fill-in reaction containing 32P-dATP, 32P-dCTP (Mc Pherson et al. Nature. 2001, 409:934-941). The resulting probe is a high specific activity 40mer which is used for hybridization to the nylon filters. Positive clone(s) identified will be provided as glycerol stocks. We can also provide additional services including clone DNA preparation, subcloning and shotgun sequencing, etc.

What you get

At the completion of the project you are provided with positive clone(s) in glycerol stock(s).  A complete report of screening results is also provided.
